We need to start a massive research effort to design life to live for extended times in space without the technology. Cause its the technology that breaks down.
Step 0: Establish a space-survival genomics research initiative, with special exclusions from bioethics regulations (not all, but perhaps some - trying to learn something here).
Step 1: Create a photosynthetic mouse. Try it. See if it results in a decreased infrastructure requirement for space habitation.
Step 2: Create a photosynthetic chimp.
Step 3: Try other modifications for heat management.
Step 4: Find protein synthesis pathways that can use a nuclear thermal battery as an energy source.
Step 5: Survival in a vacuum.
Step 6: Bioelectric radio signaling
Step 7: Chromataphor visual communication
Step 8: Direct chromataphor electric machine interface
Step 9: Optical polarization biosensors
Step 10: Biological optical interferometers
... and on up the evolutionary ladder. Per Ardua Ad Astra (through adversity to the stars).

I think the technology needed to sustain life in space is the problem
