A single Shuttle mission runs closer to a billion dollars. Launching everything you need for a space mission from the Earth's surface is very expensive. So, for that matter, is establishing infrastructure in space. However, once the infrastructure is in place, things get a lot cheaper.
Infrastructure changes everything. Time was when it took weeks to months to drive across the US. The roads, such as they were, were very poor. there weren't very many bridges. There weren't many places to buy gasoline or parts. Fortunately, some people decided that building the infrastructure for the highway system was a good idea. They didn't just look at the way things were and say "baloney". They had vision, and today you can drive across the country in a few days without strain. That highway system has become one of the major backbones for our economy.

A Titan IV-Centaur launch costs about $400 million
